Im 76 and used to have back pain until My Grandfather told me to "Drink Water"...My Mother said to Drink water to get rid of back pains....I still drink water...usually 3-5 litres of well water every 24 hours. and no more back pain....I have passed this info to my Kids..and they don't have back pain either....the dozens I have told to drink well; water 3-5 a day......all seem to be pain free..humm...

not pop or tea or coffee as water....just WATER.....but I guess its 'more convenient' to swallow a few pain pills.....and real ASA...not the garbage like Tylenol and the hundreds of other toxic chemicals............IDK I guess most don't seem to care what they gobble down as nourishment....or even what they pump into their bodies just to 'feel good' what ever that is.......

Cardiolipin (CL) plays an essential role in maintaining mitochondrial ultrastructure and function in multiple cell types. Aberrant CL metabolism has been directly linked to a number of physiological and developmental abnormalities, including embryonic lethality, cardiomyopathy, immune cell defects, hematopoietic stem cell dysfunction, neuronal dysfunction, and metabolic disorders.

CL is crucial for regulating various mitochondrial functions, ranging from respiration and metabolism to apoptosis. CL may also play an important role in regulating mitochondrial metabolism. It has been shown that CL can interact with several mitochondrial transporter proteins. The reduction in CL levels precedes losses in other phospholipids.

Mitochondrial functions in sensory neurons reduce hyperalgesia in preclinical models of neuropathic and inflammatory pain. Genetic alteration of complex IV of the mitochondrial electron transport chain in primary sensory neurons causes hypersensitivity to pain. Furthermore, 70% of humans with inherited mitochondrial deficits develop chronic pain. These data suggest an association between mitochondrial dysfunction, neuronal activity and chronic pain. In table 1 and 2 of the first link, a detailed description of the preclinical findings that show mitochondrial dysfunction in inflammatory and neuropathic pain models. Mitochondria generate approximately 90% of cellular ROS. MtROS production damages mitochondria in a variety of pathologies, including neuropathies. MtROS causes translocation of the protein cardiolipin from the inner mitochondrial membrane to the outer mitochondrial membrane, where cardiolipin serves as a docking site for caspase-1 and NLRP3. Activation of the neuronal NLRP3 inflammasome likely contributes to neuroinflammation and neurodegeneration.

Systemic or intrathecal administration of nicotinamide riboside, a precursor of NAD+, reversed inflammatory pain. Importantly, NAD+ supplementation with NAD+ precursors, such as nicotinamide riboside, mitigates oxidative stress and improves mitochondrial functions. Studies have already shown that NAD+ supplementation reduces pain.

The antioxidant resveratrol is effective in the treatment of rheumatoid arthritis by reducing plasma inflammatory markers and joint inflammation. Oral resveratrol also attenuated chronic musculoskeletal pain https://www.ncbi.nlm.nih.gov/pmc/articles/PMC9619239/ (2022).—

Through the positive regulation of Nrf2 and the anti-inflammatory effect partly through the inhibition of NF-κB activation. DHA has been shown to readily replace linoleic acid in mitochondrial cardiolipin, resulting in a change in mitochondrial respiration rate and modulation of mitochondrial-dependent apoptosis. EPA and DHA increase mitochondrial membrane potential and cardiolipin levels.

Several studies on the association of curcumin with lipid assemblies have shown that the molecule positively affects the structure and dynamics of lipids and perturbs membrane properties. Furthermore, curcumin rapidly accumulates in the endoplasmic reticulum (ER) and lysosomes, impacting both ER stress and mitochondrial functional pathways.
