The potential of vitamin D for cancer prevention and adjunctive treatment has long been bolstered by several discovered mechanisms involving the regulation of cell growth and differentiation, apoptosis, intercellular contacts, angiogenesis, immune function, and interaction with the gut microbiome. Calcitriol induces the transcription of several pro-apoptotic genes, which encode or interact with Bcl-2 family proteins in ways that enhance mitochondrial membrane permeability and apoptosis. Rapid cell growth and extracellular matrix disruption are addressed by tumor cells with pro-vascularization signaling via VEGF, angiopoietin-1, and platelet-derived growth factor (PDGF) to meet their oxygen needs. An anti-angiogenetic effect of calcitriol was supported in human cancer cells and mice, in which calcitriol decreased VEGF expression and tumor vascularization was observed. Table 1.
